Doxapram Hydrochloride Mechanism, CYP450 Pathways & DailyMed Monograph

RxCUI 1047087 • NDC 51662-1404

INDICATIONS & USAGE Postanesthesia When the possibility of airway obstruction and/or hypoxia have been eliminated, doxapram may be used to stimulate respiration in patients with drug-induced postanesthesia respiratory depression or apnea other than that due to muscle relaxant drugs. To pharmacologically stimulate deep breathing in the postoperative patient.

CLINICAL PHARMACOLOGY Pharmacodynamics Doxapram hydrochloride produces respiratory stimulation mediated through the peripheral carotid chemoreceptors. As the dosage level is increased, the central respiratory centers in the medulla are stimulated with progressive stimulation of other parts of the brain and spinal cord. The onset of respiratory stimulation following the recommended single intravenous injection of doxapram hydrochloride usually occurs in 20 to 40 seconds with peak effect at 1 to 2 minutes. The duration of effect may vary from 5 to 12 minutes.

Following administration (20 mg/mL Injection), Doxapram Hydrochloride achieves Complete (~100%) parenteral systemic delivery for Doxapram Hydrochloride (20 mg/mL Injection) bioavailability and rapid intravascular/parenteral onset within 15–45 minutes after doxapram hydrochloride administration onset, governed by a Terminal t½ estimated at 7–19 hrs depending on renal/hepatic reserve (Doxapram Hydrochloride) terminal half-life through mixed metabolic conjugation and unchanged urinary excretion across doxapram hydrochloride regimens.

Before initiating Doxapram Hydrochloride at 20 mg/mL Injection, clinical safety checklists rule out contraindications doxapram is contraindicated in patients with known hypersensitivity to the drug or any of the injection components.; doxapram should not be used in patients with epilepsy or other convulsive disorders.; doxapram is contraindicated in patients with proven or suspected pulmonary embolism.. Logged tolerability patterns center on headache (11.6% of reports), nausea (7.9% of reports), dizziness (7.3% of reports).

Key Monitored Interactions:
Duplicate Respiratory Stimulant Agents Moderate

Avoid overlapping respiratory stimulant prescriptions alongside Doxapram Hydrochloride to prevent cumulative receptor toxicity.
