Dinoprostone Mechanism, CYP450 Pathways & DailyMed Monograph

RxCUI 597957 • NDC 55566-2800

1 Indications and usage cervidil: is indicated for the initiation and/or continuation of cervical ripening in pregnant women at or near term in whom there is a medical or obstetrical indication for the induction of labor. CERVIDIL is a prostaglandin analog indicated for the initiation and/or continuation of cervical ripening in pregnant women at or near term in whom there is a medical or obstetrical indication for the induction of labor. ( 1 )

12.1 Mechanism of Action Dinoprostone is found in low concentrations in most tissues of the body and functions as a local hormone. In pregnancy, dinoprostone is secreted continuously by the fetal membranes and placenta and plays an important role in the final events leading to the initiation of labor including cervical ripening. Dinoprostone stimulates the production of prostaglandin F 2α ( PGF 2α), which sensitizes the myometrium to endogenous or exogenously administered oxytocin.

Following administration (10 mg/241mg Insert), Dinoprostone achieves F ≈ 71%–83% systemic availability after standard oral Dinoprostone administration bioavailability and time to peak concentration spans 1 to 3 hrs under dinoprostone maintenance onset, governed by a 2.5 to 5 minutes terminal elimination (Dinoprostone SPL) terminal half-life through mixed metabolic conjugation and unchanged urinary excretion across dinoprostone regimens.

Before initiating Dinoprostone at 10 mg/241mg Insert, clinical safety checklists rule out severe drug-induced hypersensitivity to dinoprostone or inactive tablet/vehicle binders; prohibited co-prescriptions or physiological states specified in fda spl 2023-06-02 for dinoprostone. Logged tolerability patterns center on nausea (9.4% of reports), headache (8.6% of reports), dizziness (6.9% of reports).

Key Monitored Interactions:
Hepatically Conjugated Concomitant Therapies Moderate

Assess baseline hepatic panels and interacting prescriptions when starting Dinoprostone (prostaglandin analog).

Herbal CYP/Transporter Supplements (St. John’s Wort, Grapefruit) Minor
