Latanoprostene Bunod Mechanism, CYP450 Pathways & DailyMed Monograph

RxCUI 1988394 • NDC 24208-504

1 Indications and usage vyzulta: ® (latanoprostene bunod ophthalmic solution) 0.024% is indicated for the reduction of intraocular pressure (IOP) in patients with open-angle glaucoma or ocular hypertension. VYZULTA is a prostaglandin analog indicated for the reduction of intraocular pressure in patients with open-angle glaucoma or ocular hypertension. ( 1 )

12.1 Mechanism of Action Latanoprostene bunod is thought to lower intraocular pressure by increasing outflow of aqueous humor through both the trabecular meshwork and uveoscleral routes. Intraocular pressure is a major modifiable risk factor for glaucoma progression. Reduction of intraocular pressure reduces risk of glaucomatous visual field loss.

Systemic exposure to Latanoprostene Bunod reflects Oral fraction absorbed ranges from 56% to 68% (Latanoprostene Bunod .24) absorption kinetics and dual hepatorenal clearance pathway combining glucuronidation and urinary filtration of latanoprostene bunod, sustaining receptor engagement across a Mean elimination half-life of 7 to 17 hours across adult Latanoprostene Bunod dosing half-life window (oral tmax achieved within 2 to 5 hours following latanoprostene bunod ingestion response profile).
