12.1 Mechanism of Action Naproxen has analgesic, anti-inflammatory, and antipyretic properties. Naproxen sodium has been developed as a more rapidly absorbed formulation of naproxen for use as an analgesic. The mechanism of action of naproxen, like that of other NSAIDs, is not completely understood but involves inhibition of cyclooxygenase (COX-1 and COX-2). Naproxen is a potent inhibitor of prostaglandin synthesis in vitro. Naproxen concentrations reached during therapy have produced in vivo effects. Prostaglandins sensitize afferent nerves and potentiate the action of bradykinin in inducing pain in animal models.

Following administration (550 mg/1 Tablet, Film Coated), Naproxen Sodium achieves ~95% systemic bioavailability (Naproxen Sodium) bioavailability and initial pharmacodynamic onset within 1–3 hrs (peak cmax) for naproxen sodium onset, governed by a Biphasic clearance with 11–18 hr terminal t½ under Naproxen Sodium maintenance terminal half-life through hepatic enzymatic processing and renal clearance proportional to creatinine clearance (naproxen sodium).

Before initiating Naproxen Sodium at 550 mg/1 Tablet, Film Coated, clinical safety checklists rule out naproxen tablets and naproxen sodium tablets are contraindicated in the following patients:; known hypersensitivity (e.g., anaphylactic reactions and serious skin reactions) to naproxen or any components of the drug product; history of asthma, urticaria, or other allergic-type reactions after taking aspirin or other nsaids.. Logged tolerability patterns center on nausea (9.8% of reports), headache (8.4% of reports), abdominal discomfort (6.2% of reports).

Key Monitored Interactions:
NSAIDs & Salicylates (Ibuprofen, Naproxen, Aspirin) Moderate

Pairing Naproxen Sodium with chronic NSAIDs can blunt renal prostaglandin perfusion or irritate gastric mucosa.

Duplicate Nonsteroidal Anti-inflammatory Drug Agents Moderate

Avoid overlapping nonsteroidal anti-inflammatory drug prescriptions alongside Naproxen Sodium to prevent cumulative receptor toxicity.
